A Peanut Variety ‘Heuksaeng’ with Short Stem and Purple Testa

  • A new peanut variety ‘Heuksaeng’(Arachis hypogaea ssp. hypogaea L.) was developed at the Department of Southern Area Crop Science, National Institute of Crop Science (NICS) in 2014. This was developed from the cross between ‘Iksan 31’ with Virginia typed short stem and ‘Iksan 35’ with large grain. ‘Heuksaeng’ is the first purple testa variety developed in Korean. It has 25 branches per plant and its length of main stem was 32 cm. Each pod has two grains with puple testa and long ellipse-shaped kernel. Its yield components showed 60 pods per plant, 69 g of 100-seed-weight and 77% of pod shelling ratio in the regional yield trials (RYT). Seed quality showed 46.0% of crude oil and 26.9% of protein content. This variety has anthocyanins typed as delpinidin-3-glucoside (D3G) and cyanidin-3-glucoside (C3G) in seed testa. This variety showed more resistant to late leaf spot, web blotch, and lodging, compared to reference variety. In the regional yield trials for 2 years, ‘Heuksaeng’ with 4.25 MT/ha for grain production has the same productivity to reference variety.

최근 건강기능성이 좋은 것으로 알려진 견과류가 국민의 간식 거리가 되면서 외국으로부터 수입이 매년 확대되어 연간 3,300 억원에 이르고 있다. 수입액에서 아몬드가 47%로서 가장 높고, 호두 23%, 밤 16%, 케슈넛 4%, 파스타지오 2% 순이다(품질관 리원 자료 2012). 수입액에 포함되지 않았던 땅콩까지 더하면 수입액은 훨씬 증가할 것이다. 땅콩은 이들 견과류와 경쟁관계로 고품질 땅콩 개발은 시급히 요구되고 있다. 땅콩은 유지작물로서 양질의 기름함량이 약 50%이며, 단백질도 25%나 함유하고 각종 비타민과 무기성분을 다량 함유하여 영양면에서도 우수하며, 광합성 능력도 높아 두과작물 중 수량이 높은 편이다(Pattee & Stalker 1995). 세계적인 땅콩 이용 형태는 착유용, 가공용 또는 간식용으로 이용되고 있으나(Park et al. 1995), 국내소비는 대립종 위주의 간식용과 소립종 위주의 제과와 제빵 같은 가공식 품용으로 수요가 크게 구분 되고 있는 실정이다(Lee et al. 1989, Park et al. 1995). 최근에는 기능성성분 레스베라트롤 (resveratrol) 함량이 높은 땅콩 싹나물 개발(Pae et al. 2011)과 기름의 올레산 함량이 80%이상으로 산화안정성이 높은 ‘케이 올’ 땅콩 품종 개발(Yang et al. 2010, Pae et al. 2016), 안토시아 닌 함유 검정땅콩도 개발 중으로 국산 땅콩 차별화에 힘쓰고 있다. 땅콩 종피의 안토시아닌 연구는 아직 미흡하여 땅콩 종피색 이 다른 22종에서 그 중 anthocyanin함유 검정땅콩 4종에 대해 cyanidin-3-glucoside을 조사하였을 정도이다(Yoav et al. 2012). 상대적으로 검정콩의 안토시아닌 연구는 활발하여 품종 별 함량, 안토시아닌 종류, 및 그 생리 활성이 진행되고 있다 (Choung et al. 2001).
우리나라는 열대원산인 땅콩의 재배 북한계기에 속해 그 동안 기상과 소비자 기호에 맞는 조숙 소분지 대립 다수성 신풍 초형을 개발함과 동시에 용도별 다양한 품종을 개발하여 왔다. 비닐피복 재배를 통하여 파종기를 앞당기게 됨으로써 대립종 재배가 가능 하였고 단위 면적당 수량을 높이는 재배법을 개발하였다 (Lee et al. 1983, Lee et al. 1986, Lee et al. 1987, Lee et al. 1989). 또한 작부체계에 알맞은 풋땅콩 품종개발 및 노력이 많은 수확작 업의 생력기계화도 보고 되고 있다(Kim et al. 1990, Pae et al. 2002, Pae et al. 2007).
국립식량과학원 남부작물부에서 2004부터 땅콩연구를 전담 하게 되어 그 동안 대립 다수성 땅콩 품종을 육성하기 위하여 노력하였다. 그 결과 초형을 단경화하여 도복에 강하고 기계화 수확에 유리한 다분지 버지니아 초형 품종 ‘상평’, ‘풍안’ 등을 육성 하였으며(Pae et al. 2009), 이번에는 단경 반직립 초형으로 종실의 크기가 중립종이며 성숙이 잘 되는 안토시아닌 함유 검정땅콩 ‘흑생’을 국내최초로 육성하게 되었기에 이 품종의 육성경위와 주요특성을 요약 보고하는 바이다.
신품종 ‘흑생’(밀양61호)의 특성 및 수량검정 시험은 2013부터 2014까지 2년간 4개 지역(청원, 익산, 대구, 밀양)에서 지역적응시 험을 수행하여 표준품종 ‘대광’땅콩과 3반복으로 비교 검토하였 다(RDA 2014). 시비량은 ha당 N-P2O5-K2O로서 30-100-100 kg을, 석회와 퇴비는 각각 1 ton과 10 ton을 전량기비로 시용하였 다. 재식거리는 조간거리 40 cm, 휴간거리 60 cm, 주간거리는 25 cm로서 1휴 2열 재배방식으로 ha당 80,000 주로 하였다. 비닐피복은 흑색유공비닐을 이용하였으며 규격은 폭 110 cm, 두께 0.015 mm, 구멍크기 5 cm였다. 파종기는 전 시험지에서 4월 30일로 2립씩 파종하여 출현 후 솎음 작업으로 1주1본으로 재배하였다. 주경장, 분지장 및 분지수는 각 시험구에서 고유특 성이 잘 나타나는 균일지역을 선정하여 10주씩 조사하였다. 수확 시기는 개화 후 95 일경에 각 시험구당 6 m2씩 굴취하여 햇볕에 충분히 풍건한 다음 협의 그물무늬가 뚜렷하고 상품가치가 높은 것을 성숙협으로 간주하여 종실수량 및 수량구성요소를 조사하 였다. 이들 조사는 땅콩지역적응시험 조사기준에 준하여 실시하 였으며(RDA 2013), 4개 지역에서 2년간 수행한 시험성적을 정리한 평균치를 품종간 생육 및 수량구성요소의 비교 값으로 활용하였다(RDA 2014). 종실 성분분석에서 기름을 Soxtec (B-811, Buchi, Switzerland) 분석법을 이용하여 20회 추출 후 측정하였고, 지방산은 GC(7890A, Agilent, USA)를 이용해 분 석하였고, 단백질은 자동 Kjeldahl(rapip N cube, Elementar, Germany)을 이용하였다. 땅콩껍질의 안토시안닌은 1g의 껍질 을 20% 메탄올과 1% 염산이 혼합된 용액 30ml에서 48시간 추출하여 HPLC(Dionix, Lichro CART RP-18e column)에서 분석하였다. 그 외 조사는 일반 분석방법(Kim et al. 1990)으로 하였다.
육성경위
‘흑생’은 국내 생산자와 소비자 선호도가 높은 검정종피 땅콩 을 육성코자 단경 내재해 다수성 품종 육성에 목표를 두고 선발 육성된 신품종으로 다분지 단경 계통인‘익산31호’를 모본으로 소 분지 검정 종피 계통인 ‘밀양35호’를 부본으로 2005년 인공교배 후 계통육성 과정을 거쳐 선발되었다. ‘밀양61호’는 2년간 생산력검 정시험(2012~2013)과 2년간 지역적응시험(2013~10214, RDA) 을 수행하였는데 조속한 검정땅콩 품종육성을 위해 2013년에는 위 두 시험을 동시에 수행하기도 하였다 (Fig. 1). 그 결과 다분지 단경 다수성인 ‘밀양61호’는 검정땅콩 품종으로 우수성이 인정되 어 2014년 12월 농촌진흥청 신품종선정위원회에서 신품종으로 선정하고 대국민 품종명 공모에 의하여 ‘흑생’으로 명명하였다.

고유특성
‘흑생’의 고유 특성은 대비 품종인 ‘대광’ 땅콩 (Lee et al. 1986)의 신풍 초형과 달리 분지수가 많은 버지니아 초형으로 주경에서만 착협한다. 협당립수는 2립이고, 잎은 연녹색이며 크기가 작으고, 종피색은 검정색, 종실형태는 장타원형으로 알이 더 작은 중립종에 속한다(Table 1).

일반특성
지상부 생육특성으로 ‘흑생’의 개화기는 6월 7일로서 ‘대광’ 보다 1일 늦었다. 분지수가 적은 초형의 품종이 가지가 많은 초형의 품종 보다 개화가 빠른 특성(Lee et al. 1983)을 보이는 것이 일반적이다. 주경장은 32 cm, 분지장은 40cm로서 ‘대광’땅 콩에 비해 각각 20cm, 27cm 더 짧고, 분지수가 25개인 다분지 단경 초형 특성을 나타낸다(Table 2). 기존의 직립성인 초형과는 달리 반직립성 다분지 초형이므로 초기와 중기에서 초관 형성이 좋고 후기에 성숙을 잘하는 특성을 가지고 있음을 알 수 있다.

지하부 특성으로 ‘흑생’은 수량구성요소 면에서 주당 성숙협 수가 60개로서 ‘대광’보다 14개나 많으나, 협당립수가 1.72개, 100립중이 69g으로 ‘대광’땅콩에 비해 낮은 경향을 보이고 있으 며, 성숙성에 있어서는 성숙협비율(87%)과 협실비율(77%)이 높아 성숙이 잘 됨을 보여준다(Fig. 2, Table 3).

내재해성
‘흑생’의 육성지인 밀양에서 정밀한 조사를 위해 따로 포장 병해검정시험을 2년간 실시하여 등숙기와 수확기에 병반면적율 을 달관 조사한 평균치의 결과, ‘흑생’의 주요 병해 저항성으로서 낙엽병류인 갈반병(Cercospora arachidicola)은 ‘대광’에 비해 약간 발생하였고 흑반병(Cercospodium personatum)과 그물무 늬병(Phoma arachidicola)은 약간 강한 특성을 보이고 있고, 생육후기에 발생하여 포기째 고사하여 수량에 영향이 큰 흰비단 병(Sclerotium. rolfsii)은 ‘대광’과 같은 수준이다. ‘흑생’은 반직 립 단경 초형이므로 후기 도복에는 강한 특성을 보이고 있다. 잎 피해로 문제가 되는 담배거세미등에 대한 충해저항성은 같은 수준이다(Table 4).

품질특성
‘흑생’땅콩의 종실 조단백 함량은 26.9%, 조지방 함량은 46.0%로서 ‘대광’에 비해 각각 1.2%, 4.6% 낮은 수준이다. 지방산 조성면에서 양질불포화 지방산인 올레산은 53.4%로서 ‘대광’에 비해 5.0% 낮으나, 리놀산이 31.2%로서 5.1% 더 높은 특성을 나타내고 있다. 미국 땅콩 유전자원에서 단백질 함량은 16.2~36% 분포를 보이며 평균 25%, 지방 함량은 35.5~54.2% 에 분포하며 평균 약 45%라 하였다(Pattee & Stalker 1995). 일반적으로 국내 땅콩 지방산 중 올레산 함량은 42~59%에 분포 하였으며 올레산은 리놀과 부의 상관관계를 나타내고 있다(Kim et al. 1990, Lee et al. 1989). ‘흑생’ 종피의 안토시아닌은 Delpinidin-3-glucoside(D3G)와 Cyanidin-3-glucoside(C3C) 등 2종류를 가지며 합계 58.5ug/g을 함유하고 이었다(Table 5). ‘흑생’은 Cyanidin-3-glucoside(C3C) 1종만 함유한 기존 문헌(Yoav et al. 2012)과는 다른 양상을 보였다.

수량성
2012년부터 2013년까지 2개년간 실시한 생산력검정시험(밀 양) 결과 ‘흑생’의 ha당 평균 종실수량은 3.95ton으로 ‘대광’땅콩 대비 11%높았다(Table 6).

2013년부터 2014년까지 2개년간 전국 4개 지역에서 실시된 지역적응시험 결과 ‘흑생’의 ha당 평균 수량성은 4.25ton으로 ‘대광’ 땅콩과 같은 수준으로 유의성이 인정되지 않았다. ‘흑생’ 땅콩은 국내최초로 개발된 검정종피 땅콩으로 단경 다분지 초형 으로 개량됨에 따라 ‘대광’에서 문제시 되었던 성숙 후기 도복과 병해가 개선되어 협수, 협당립수, 100립중 등 수량구성요소가 양호하게 되에 중립종에도 불구하고 ‘대광’ 수준의 수량성을 발휘한 것으로 생각된다(Table 7).

적응지역 및 재배상의 유의점
‘흑생’ 땅콩은 중립종으로 경기북부와 강원도 산간지를 제외 한 전국 평야지역에서 재배가 가능한 품종이다. 재배상 유의점으 로는 초기의 생육과 개화수를 충분히 확보하고 성숙기의 적습을 통하여 자방병 지하침투와 착과성을 원활히 하며, 후기 영양결핍 으로 잎 끝의 황화가 오지 않도록 유의해야 하며, 병해 및 충해 방제를 철저히 하여 등숙성을 좋게 하면 수량 특성을 잘 발휘할 수 있다. 특히 수확시기가 늦어지면 종피의 검정색이 탈색되어 품질이 저하 되거나 꼬투리에서 수발아 발생할 수 있으므로 개화기 이후 90일이내에 수확이 되어야 한다.
기능성 땅콩 선호도가 높음에 따라 국내최초 안토시아닌 함유 검정종피 신품종 육성을 위하여 단경 다분지 계통인 ‘밀양31호' 와 소분지 검정계통인 ‘밀양35호’을 인공교배하여 육성한 ‘흑생’ 의 주요 특성을 요약하면 다음과 같다. ‘흑생’은 버지니아 초형으 로 개화기가 ‘대광’에 비해 1일 늦으나 지상부 생장습성은 다분지 단경 반직립종이다. ‘흑생’은 수량구성요소 면에서 주당 성숙협 수는 60개로서 ‘대광’ 땅콩에 비해 14개 많으나 협당립수(1.72 개), 100립중(69g) 등 은 약간 낮은 중립종이며, 성숙협비율 (87%)과 협실비율(77%)이 높아 후기 성숙성이 좋은 박피종이 다. 병해저항성으로 흑반병과 그물무늬병도 ‘대광’에 비해 다소 강한 편이이며 도복은 강한 편이다. ‘흑생’은 단백질이 26.9%, 기름함량이 46%이고 지방산비율은 양질지방산인 올레산이 53.4%, 리놀산이 31.2%인 특성을 가지고 있다. ‘흑생’은 2년간 지역적응시험 결과 전국 평균 4.25ton로서 ‘대광’과 동일한 수준 이다. 재배상 유의 점으로는 성숙기 포장의 수분관리와 흑반병과 그물무늬병 등 병해 방제에 유의 해야 꼬투리의 알이 잘 성숙한 다. 특히 수확시기는 개화 후 90일을 넘기지 않도록 해야 하며 전국평야지 지역에서 재배가 가능한 품종이다.
